Another development of Google threatening Apple is their new design of home theater systems of Televisions partnering with Sony and Samsung Companies.               The second critical external factor affecting Apple Company is continued international legal disputes around the world. Apple being the leading innovators for their line of products makes them be the hunted and not the hunters. One of their going international legal disputes is the ongoing co-wireless licensing patent lawsuit. Co-wireless licensing patent an ally of Microsoft is suing Apple for wireless patents. If the lawsuit were to go through, this would be a huge blow for Apple since most of their products operate on a wireless network.                Apple is also facing another international legal dispute following claims that Apple purchased iPad trademark illegally. With their distinctive feature of their products starting with an ‘i’, this lawsuit could turn out to be a catastrophe for the company disrupting the flow of Apple tablet market share. These legal disputes are bad for business for the Apple Company and if they were to go through it could mark the end of the leading innovators Apple INC.                  Stakeholders in an organization include customers, employees, the entire community, suppliers and the business partners. Customers and the community are the key stakeholders in the success of an organization. Satisfied customers and the whole community lead to real business. Primary stakeholders affect an organization mostly financially. Primary stakeholders include shareholders, employees, and suppliers.                Primary stakeholders contribute to an organization’s environmental risks that impact negatively on financial performance of an organization. Steve Jobs exact pressure on his employees and many consider him a tough leader; as Apple continues to record profits in their financial reports.                Another factor that affects financial performance of an organization is the structure and working of an organization. Apple Company has a well-structured supply chain management that coordinates its suppliers, the operations of the organization and the customers. This coordination by Apple and its business partners ensures smooth operations in the Company eliminating possibility of registering loses.                Labor practices are a controversial corporate social responsibility concern associated with Apple. Apple has been accused of overworking its’ employees following recent speculations from two supplier shops in China known as Foxconn and Inventec. Media recorded that; workers in these Companies are forced to work long hours and are underpaid. In addition, they are forced to live on the premises of these Companies where they have to pay rent and other expenses from the little they earn from the Company. This has raised debates over Chinese labor regulations and whether it is entirely Apple’s fault of Foxconn fault. Current Status of Gene Therapy Research

Current Status of Gene Therapy Research Genes, which are carried on chromosomes, are the basic physical and functional units of heredity. Genes are specific sequences of bases that encode instructions on how to make proteins. Although genes get a lot of attention, its the proteins that perform most life functions and even make up the majority of cellular structures. When genes are altered so that the encoded proteins are unable to carry out their normal functions, genetic disorders can result. Gene therapy is a technique for correcting defective genes responsible for disease development. Researchers may use one of several approaches for correcting faulty genes: A normal gene may be inserted into a nonspecific location within the genome to replace a nonfunctional gene. This approach is most common. An abnormal gene could be swapped for a normal gene through homologous recombination. The abnormal gene could be repaired through selective reverse mutation, which returns the gene to its normal function. The regulation (the degree to which a gene is turned on or off) of a particular gene could be altered. How does gene therapy work? In most gene therapy studies, a normal gene is inserted into the genome to replace an abnormal, disease-causing gene. A carrier molecule called a vector must be used to deliver the therapeutic gene to the patients target cells. Currently, the most common vector is a virus that has been genetically altered to carry normal human DNA. Viruses have evolved a way of encapsulating and delivering their genes to human cells in a pathogenic manner. Scientists have tried to take advantage of this capability and manipulate the virus genome to remove disease-causing genes and insert therapeutic genes. Target cells such as the patients liver or lung cells are infected with the viral vector. The vector then unloads its genetic material containing the therapeutic human gene into the target cell. The generation of a functional protein product from the therapeutic gene restores the target cell to a normal state. See adiagram  depicting this process. Some of the different types of viruses used as gene therapy vectors: Retroviruses   A class of viruses that can create double-stranded DNA copies of their RNA genomes. These copies of its genome can be integrated into the chromosomes of host cells. Human immunodeficiency virus (HIV) is a retrovirus. Adenoviruses   A class of viruses with double-stranded DNA genomes that cause respiratory, intestinal, and eye infections in humans. The virus that causes the common cold is an adenovirus. Adeno-associated viruses   A class of small, single-stranded DNA viruses that can insert their genetic material at a specific site on chromosome 19. Herpes simplex viruses   A class of double-stranded DNA viruses that infect a particular cell type, neurons. Herpes simplex virus type 1 is a common human pathogen that causes cold sores. Besides virus-mediated gene-delivery systems, there are several nonviral options for gene delivery. The simplest method is the direct introduction of therapeutic DNA into target cells. This approach is limited in its application because it can be used only with certain tissues and requires large amounts of DNA. Another nonviral approach involves the creation of an artificial lipid sphere with an aqueous core. This liposome, which carries the therapeutic DNA, is capable of passing the DNA through the target cells membrane. Therapeutic DNA also can get inside target cells by chemically linking the DNA to a molecule that will bind to special cell receptors. Once bound to these receptors, the therapeutic DNA constructs are engulfed by the cell membrane and passed into the interior of the target cell. This delivery system tends to be less effective than other options. Researchers also are experimenting with introducing a 47th (artificial human) chromosome into target cells. This chromosome would exist autonomously alongside the standard 46 not affecting their workings or causing any mutations. It would be a large vector capable of carrying substantial amounts of genetic code, and scientists anticipate that, because of its construction and autonomy, the bodys immune systems would not attack it. A problem with this potential method is the difficulty in delivering such a large molecule to the nucleus of a target cell. What is the current status of gene therapy research? The Food and Drug Administration (FDA) has not yet approved any human gene therapy product for sale. Current gene therapy is experimental and has not proven very successful in clinical trials. Little progress has been made since the first gene therapy clinical trial began in 1990. In 1999, gene therapy suffered a major setback with the death of 18-year-old Jesse Gelsinger. Jesse was participating in a gene therapy trial for ornithine transcarboxylase deficiency (OTCD). He died from multiple organ failures 4 days after starting the treatment. His death is believed to have been triggered by a severe immune response to the adenovirus carrier. Another major blow came in January 2003, when the FDA placed a temporary halt on all gene therapy trials using retroviral vectors in blood stem cells. FDA took this action after it learned that a second child treated in a French gene therapy trial had developed a leukemia-like condition. Both this child and another who had developed a similar condition in August 2002 had been successfully treated by gene therapy for X-linked severe combined immunodeficiency disease (X-SCID), also known as bubble baby syndrome. FDAs Biological Response Modifiers Advisory Committee (BRMAC) met at the end of February 2003 to discuss possible measures that could allow a number of retroviral gene therapy trials for treatment of life-threatening diseases to proceed with appropriate safeguards. In April of 2003 the FDA eased the ban on gene therapy trials using retroviral vectors in blood stem cells. What factors have kept gene therapy from becoming an effective treatment for genetic disease? Short-lived nature of gene therapy   Before gene therapy can become a permanent cure for any condition, the therapeutic DNA introduced into target cells must remain functional and the cells containing the therapeutic DNA must be long-lived and stable. Problems with integrating therapeutic DNA into the genome and the rapidly dividing nature of many cells prevent gene therapy from achieving any long-term benefits. Patients will have to undergo multiple rounds of gene therapy. Immune response   Anytime a foreign object is introduced into human tissues, the immune system is designed to attack the invader. The risk of stimulating the immune system in a way that reduces gene therapy effectiveness is always a potential risk. Furthermore, the immune systems enhanced response to invaders it has seen before makes it difficult for gene therapy to be repeated in patients. Problems with viral vectors   Viruses, while the carrier of choice in most gene therapy studies, present a variety of potential problems to the patient toxicity, immune and inflammatory responses, and gene control and targeting issues. In addition, there is always the fear that the viral vector, once inside the patient, may recover its ability to cause disease. Multigene disorders   Conditions or disorders that arise from mutations in a single gene are the best candidates for gene therapy. Unfortunately, some the most commonly occurring disorders, such as heart disease, high blood pressure, Alzheimers disease, arthritis, and diabetes, are caused by the combined effects of variations in many genes. Multigene or multifactorial disorders such as these would be especially difficult to treat effectively using gene therapy. The School of Pharmacy in London is testing a treatment in mice, which delivers genes wrapped in nanoparticles to cancer cells to target and destroy hard-to-reach cancer cells. Read  BBC article. Results of worlds first gene therapy for inherited blindness show sight improvement. 28 April 2008. UK researchers from the UCL Institute of Ophthalmology and Moorfields Eye Hospital NIHR Biomedical Research Centre have announced results from the worlds first clinical trial to test a revolutionary gene therapy treatment for a type of inherited blindness. The results, published today in the New England Journal of Medicine, show that the experimental treatment is safe and can improve sight. The findings are a landmark for gene therapy technology and could have a significant impact on future treatments for eye disease. Read  Press Release. Previous information on this trial (May 1, 2007): A team of British doctors from Moorfields Eye Hospital and University College in London conduct first human gene therapy trials to treat Lebers congenital amaurosis, a type of inherited childhood blindness caused by a single abnormal gene. The procedure has already been successful at restoring vision for dogs. This is the first trial to use gene therapy in an operation to treat blindness in humans. Researchers at the National Cancer Institute (NCI), part of the National Institutes of Health, successfully reengineer immune cells, called lymphocytes, to target and attack cancer cells in patients with advanced metastatic melanoma. This is the first time that gene therapy is used to successfully treat cancer in humans. Myeloid disorders are common and include a variety of bone marrow failure syndromes, such as acute myeloid leukemia. The study is the first to show that gene therapy can cure diseases of the myeloid system. Each animal had been deafened by destruction of the hair cells in the cochlea that translate sound vibrations into nerve signals. A gene, called  Atoh1,  which stimulates the hair cells growth, was delivered to the cochlea by an adenovirus. The genes triggered re-growth of the hair cells and many of the animals regained up to 80% of their original hearing thresholds. This study, which many pave the way to human trials of the gene, is the first to show that gene therapy can repair deafness in animals. University of California, Los Angeles, research team gets genes into the brain using liposomes coated in a polymer call polyethylene glycol (PEG). The transfer of genes into the brain is a significant achievement because viral vectors are too big to get across the blood-brain barrier. This method has potential for treating Parkinsons disease. Short pieces of double-stranded RNA (short, interfering RNAs or siRNAs) are used by cells to degrade RNA of a particular sequence. If a siRNA is designed to match the RNA copied from a faulty gene, then the abnormal protein product of that gene will not be produced. Some Questions to Consider What is normal and what is a disability or disorder, and who decides?   Are disabilities diseases? Do they need to be cured or prevented?   Does searching for a cure demean the lives of individuals presently affected by disabilities?   Is somatic gene therapy (which is done in the adult cells of persons known to have the disease) more or less ethical than germline gene therapy (which is done in egg and sperm cells and prevents the trait from being passed on to further generations)? In cases of somatic gene therapy, the procedure may have to be repeated in future generations.   Preliminary attempts at gene therapy are exorbitantly expensive. Who will have access to these therapies? Who will pay for their use?

In his novel The Europeans, Henry James tells the story of an American family that is visited by their European cousins. James uses these circumstances to depict the differences between Europeans and Americans. The Americans tend to be frightened of the Europeans, since they seem quite foreign within the puritanical American community. On the other hand, the Europeans are surprised by the Americans' provincial ways. Reaction to the unfamiliar is a central element of the novel. Each character's reaction to the unfamiliar reveals his or her personality and also determines whom that character is capable of tolerating and of loving. Felix, one of the European cousins, and Gertrude, the younger daughter of the American family, both illustrate characters who welcome the unknown. Charlotte, the elder daughter of the American family, and Mr. Brand, the clergyman who lives with the family, both seek to avoid change. Eugenia, the other European cousin, enjoys stability whereas her suitor Mr. Acton is excited by new experiences. The latter pair can never live together, while the characters of similar dispositions tend to become romantically involved. Thus, reaction to the unfamiliar determines the course of each of these characters' lives. Characters like Gertrude and Felix are curious about things that they have not experienced. They search for new things, being unable to maintain one way of life for a long period of time. Gertrude displays this by her imaginative nature. She adores imagining, wishing to be somewhere else in the world. When she is left alone when everyone else goes to church Gertrude decides to transport herself to Arabia: "She possessed herself of a very obvious volume-one of the series of the Arabian Nights-and sh... ...g consistency, and thus they are conservative characters who can live together because they will be able to sustain order and routine. Eugenia and Acton can never agree on how to live, forcing them to end their relationship. In this novel, James takes a more humorous approach than in his other novels, and pokes fun at his own stiff American culture. He uses the European influence to show how narrow-minded New Englanders were at the time, and in fact James was part of this puritanical society. However, he presents a contrast with characters who believe that change is essential. Thus, through showing two juxtaposed opinions, James contrasts American conservatism with European worldliness, and how this reflects on the members of those different societies. However, as in the best Shakespearean comedy, the story ends with a marriage, and it is all much ado about nothing.

Oedipus - He tried and failed. Oedipus is one of the most famous tragic characters of Greek drama. Sophocles, a famous composer of tragic Greek drama, wrote Oedipus the King. In this drama, Oedipus is just a good king trying to help his people by punishing a criminal for a certain crime he committed. Even though he tries so hard not to do what was foretold that he would do, he ends up doing it all the same. He was innocent of the knowledge of what he was doing, but guilty of the act he commits. He is a good king in trying to help his people, without his knowing it, he is the cause of the problem he is trying to solve, and when he tries to avoid the unavoidable, he finds that he is the reason for the problem. Oedipus is a good king, just trying to save his people by removing the plague. In his attempt at trying to be the savior of his people, he condemns the man who killed the former king. He wants to punish that man because he is the reason for the plague that is harming his people. He says, "Whoever he was that killed the king may readily wish to dispatch me with his murderous hand; so helping the dead king I help myself." (161-4) What he does not realize is that he is the culprit, the reason for the problem, and because he is going unpunished, the plague had continued. When Oedipus is a young adult, he goes to the oracle of Apollo, who tells him that he will one day kill his father and marry his mother. Oedipus, who has no idea that he is adopted at the time, flees his home in fear that he will cause harm to the parents he loves. After he leaves Delphi, the place of the oracle, he meets a man in a chariot whose charioteer tells him to move aside. When he refuses, the other man attacks him, and then Oedipus turns and kills them all. He comes to Thebes; the people were under siege by a Sphinx. After he saves the town, he is given the queen, Jocasta, to be his wife as a reward. What he does not know was that the one of the men he killed was his father and the woman that he marries is his mother. Oedipus the king never considers what he had done, until he hears certain pieces of information and starts to piece things together. The use of the Career Interest Profiler displayed that I was Teamwork centered portraying a work culture towards: working in a close knit team, working closely with customers and suppliers, lots of interdependence, friendly and supportive colleagues, cooperative and sharing atmosphere. The next trait of the Career Interest Profiler displayed that I was an Expert portraying a work culture towards: varied and ground breaking work, high emphasis on specialist expertise and lots of specialist roles, high engagement-people encouraged to express opinion, innovation, creative atmosphere, control your own work schedule and organization, dynamic an changing environment. The last trait of the Career interest Profiler displayed that I was Supportive portraying a work culture towards: secure, stable environment, clearly defined jobs, goals and expectation, no conflicting demands, considerate management, focus on employee welfare, lots of personal development and feedback, lots of recognition and celebration of success, and fun place to work. The Career interest Profiler reviled that I am a leader whom can work under pressure and also create an employee friendly working environment. A person who put people first and would treat employees the way I want to be treated. As a Navy Officer, Basketball player and today in my career field I’m often counted on to lead people. My approach when working with people has always been to be honest, fair and treat a person with respect and acknowledge the positive in a person because everyone will tell you when you’re doing something wrong. My Strengths in Competencies displayed I was very strong in Innovating, Coping with Pressure, and Adapting to Change. These Competencies top my measuring scale with four bars out of possible five. The Applying Expertise and Adhering to Values on the measuring scale showed two out of five possible bars and the last Competency Strength displayed Strategizing which displayed one out of five possible bars. After reviewing my Strengths in Competencies, it demonstrated to me that I can work under pressure, meet deadlines, and confirmed that I can accept change. This is true because working in my profession the Laws are always changing and the technology is always changing as well. The Work Culture Preferences displayed that I was Artistic, Conventional and Enterprising. It also showed my Career matches being Business and financial Operations, Management, Sales and Related, Legal, Community and Social Service, Personal Care and Service. This is very unique because I gave 6 years to the Military as public service, worked as a Youth Detention Officer, Youth Worker and now for the Department of Veterans Affairs as an Advocate for Veterans. The possible employers that would fit with my provided competencies would be a College, University, High school, Government, City of State Office, Police Department, Prison, Department Store, and Owner of Private business. The possible jobs that would align with my work culture preferences would be a Head basketball Coach, Detective, Police Captain, EEO Officer, and Human Resources Specialist, or Management staff. The ways in which understanding my personal competencies will help me improve my critical thinking skills is to evaluate all information I receive before making a decision on any level. This is best described for me, as receiving all the needed facts before I make any critical decision. My competencies strengths also are utilized in all efforts to help me evaluate an argument. The best example of me using my competencies strengths during an argument is when I’m engaged with a veteran who is irate, and also talking very loudly during an interview for VA benefits at my place of work. I always take control of the situation, by speaking in a much lower tone and always letting the veteran finish his statement. Most often by acknowledging the veteran and having the facts when the veteran is presenting sometimes calms them down.
